"Interferometry can be performed even with fluorescence sources. In this talk we will discuss the generation of self-interferences via a 2D phase-only diffraction grating ("Phimask"). Self-interferences obtained on single molecules fluorescence lead to single shot 3D localization of the emitter even multiple 10's of micrometer inside a (living) sample. We will discuss the implementation of Phimask on a microscope as well as the algorithm to extract the 3D position of each emitter."
